Evaluation of UV Filters in a Sunscreen for Phototoxicity Using In Vitro 3T3 Neutral Red Uptake (NRU) Phototoxicity Test

Abstract
It is well known that sunscreens protect from skin damage induced by UV. To maintain protective effect of UV filter, it`s important to keep photostability and photo-safety. According to the ban of animal test in cosmetics and its ingredients, it needs to develop the evaluating of UV filter for phototoxicity. The aim of this study was to evaluate the applicability of in vitro 3T3 NRU phototoxicity test instead of animal experiments as an alternative method. Commercially used 19 sunscreen ingredients (8 organic UV filters (O1-8), 11 mineral UV filters (M1-11)) were tested. There was no phototoxicity, in case of concentration below 1,000 §¶/§¢, however, above 1,000 §¶/§¢, two UV filters showed phototoxicity. It was estimated that the false positive took place based on existing in vivo data owing to insolubility and adhesive characteristics of ingredients. Improvements such as adjustment of concentration or ultra-sonication of chemicals are needed to apply in vitro 3T3 NRU phototoxicity test for UV filters.
